MDA-MB-231转染时，在24孔板上接种细胞密度多少合适？转染时的细胞密度达到多少合适？

大家好，小弟最近在做乳腺癌细胞MDA-MB-231转染，用的是Lipofectamine2000，小弟想咨询下在24孔板上接种细胞密度多少合适？转染时的细胞密度达到多少合适，80—90%？70%?还是别的？今天刚做了个预实验，24孔板上接种了5个密度梯度：3万/ml，5万/ml，7万/ml，10万/ml，20万/ml。不知结果怎样。。。。而Lipfectamine说明书上说（0.5-2）*100000/500微升，然后我就纠结了，大神们有做过的给点指点吧。。万分感谢。。

80% confluence
6 well是0.5-1.5 ug plasmid为佳
算一下24well吧
plasmid：2000=1:2 (M/V)

231用lipo2/3000 效果还不错，1ug质粒即可

这个细胞慢病毒感染时还是比较容易的，要看你是什么培养皿养的的才能确定你需要的病毒量

I don't think Lip2000 is good reagent for transfected the MDA 231 cell.
You don't need to follow the protocol so firm. You can increase the dose of Lip2000 and plasmid to enhance the transfection efficiency. But the base line is don't hurt the cell too much.
Anyway, you can have a try. usually in lower cell density will have higher efficiency. I test 60-70% cell density before. but the efficiency only is not more than 20%. if you only want to screen the stable cell line. it will be fine. For me, I have to change to use electroporation at last.
